Launch of Aptima HPV E6/E7 mRNA Assay

AMPATH GENETICS Laboratory and ILEX South Africa are now launching a new assay to aid in the diagnosis of host genome integration of genetic material from females who have been identified as carriers of HPV high risk genotypes. AMPATH is the first pathology group to offer this test after having being marketed heavily this week with an overseas speaker presenting data at the National Obstetrics and Gynaecology Conference in Cape Town. This assay differentiates a definitive high malignancy risk group with an integrated HPV genome, which is critical in terms of oncogenic potential, from patients carrying high risk genotypes, but with transient infections in view of absence of host integration of HPV mRNA. The latter group usually only needs a follow up investigation in about 12 months'

time. This approach decreases the "false positive" results with respect to cervical disease translating into needless anxiety as well as unnecessary medical procedures by as much as 20%.

Because HPV infections are very common and because most women will clear HPV infections within 6 to 12 months, the presence of HPV high risk DNA types does not mean that cervical dysplasia or cervical cancer is present. Studies have suggested that detection of high-risk

E6/E7 messenger ribonucleic acid (mRNA) may be a better indicator of disease progression than L1 DNA. Early genes E6 and E7 regulate HPV viral replication. The E6 and E7 genes from the pathogenic genotypes are known oncogenes and have been shown to directly contribute to malignant progression by promoting genomic instability in the patient's cells. Thus the test is a measure of host-virus genomic interaction. The APTIMA HPV E6/E7 mRNA Assay is a target amplification, nucleic acid probe test for the in vitro qualitative detection of HPV

E6/E7 viral messenger RNA (mRNA) from 14 high-risk HPV types (16/18/31/33/35/39/45/51/52/56/58/59/66/68).

The assay can be performed on either liquid-based cytological specimens or can be done using the APTIMA HPV Swab, stabilising the specimen and facilitating transport in comparison to other RNA based tests. This test is now available through AMPATH GENETICS Laboratory. Specimens can be submitted at any AMPATH depot countrywide.
